At EnXylaScope we will be doing application testing of our future products. Partner Fermentation Experts is in charge of this activity for nutraceuticals applications. Below we describe the path to be followed for this activity.
In order to investigate the Rheology and stability of the WIS-xylans an assay will be developed to analyze the sensibility towards changes in pH, Heat and addition of water. The samples will be put into petri dishes and incubated for 2 days at 37 degrees. this will both give an indication of microbiological decay and the water binding properties of the WIS-xylans produced in the EnXylascope project.
In order to investigate the microbiome modulatory properties of WIS-Xylans, the Xylans will be added to fecal samples and run through the Minigut colon model, which mimics the 24h transition time seen in the colon. Afterwards the microbiome will be characterized by 16s rRNA sequencing and analysis. Furthermore we will investigate the SCFA profile in the different samples by performing mass spectrometry.
WIS-Xylans will be incubated with Immune-assay competent model cells such as IPEC-J2, Caco2 and PBMC’s. In order to investigate the Anti-inflammatory response in the model cells, both ELISA assays and RNA sequencing will be carried out.
